Profil Penurunan Kadar Glukosa Darah Ekstrak Air Rambut Jagung (Zea Mays L.) Tua dan Muda Pada Mencit Jantan Galur Balb-C
Abstract
ABSTRAK
Rambut jagung mengandung flavonoid. Kandungan total flavonoid dipengaruhi oleh usia tanaman. Flavonoid memiliki potensi sebagai antihiperglikemia. Penelitian bertujuan untuk melihat profil penurunan kadar glukosa darah ekstrak air rambut jagung tua dan muda pada mencit jantan galur Balb-C yang diinduksi aloksan. Penelitian bersifat eksperimental dengan rancangan posttest only control group design. Ekstrak dibuat dengan merebus rambut jagung menggunakan air selama 30 menit. Total flavonoid ditentukan dengan metode AlCl3. Aktivitas penurunan kadar glukosa darah dilakukan dengan metode tes toleransi glukosa oral terhadap mencit jantan galur Balb-C yang diinduksi aloksan secara intraperitonial (140 mg/kgBB). Sebanyak 20 hewan uji dibagi menjadi kelompok kontrol positif (Akarbose 0,1305 mg/20 gBB), kontrol negatif (NaCMC 1%), ekstrak air rambut jagung tua (200 mg/kgBB) dan ekstrak air rambut jagung muda (200 mg/kgBB) yang diberi perlakuan secara oral. Kadar glukosa darah diukur pada menit ke 0, 30, 60, 90, 120, 150, dan 180 pasca pembebanan glukosa oral. Data persen penurunan kadar glukosa darah dianalisis dengan uji Kruskal Wallis dan dilanjutkan dengan uji Mann Whitney. Persen penurunan kadar glukosa darah ekstrak air rambut jagung tua dan muda tidak berbeda bermakna (p>0,10). Ekstrak air rambut jagung muda menurunkan kadar glukosa darah sejak menit ke-90 pasca pemberian glukosa oral dengan persen penuran sebesar 44,55 ± 4,92%. Sedangkan ekstrak air rambut jagung tua menurunkan kadar glukosa darah sejak menit ke-120 pasca pemberian glukosa oral dengan persen penurunan sebesar 30,15 ± 7,25%.

ABSTRACT
Corn silk contained flavonoid. Total flavonoid content could be influenced by ages of the plant. Flavonoid has potential effect as anti-hyperglycemia. The purpose of this study was to observe lowering blood glucose levels profile of mature and immature corn silk extract on male mice Balb-C strain. This is an experimental study with posttest only control group design. Aqueous extract was prepared by boiling corn silk with water for 30 minutes. Total flavonoid content was determined by AlCl3 method. Lowering blood glucose levels activity was determined by oral glucose tolerant test method on alloxan induced male Balb-C mice test animals intraperitonialy (140 mg/kgBB). The 20 test animals were divided into positive control group (Acarbose 50 mg/70kgBB), negative control group (NaCMC 1%), mature aqueous corn silk extract group (200 mg/kgBB), and immature aqueous corn silk extract group (200 mg/kgBB) given orally. Blood glucose levels were measured at 0, 30, 60, 90, 120, 150, and 180 minutes after the glucose load. Percentage of lowering blood glucose levels were analyzed by Kruskal Wallis and continued by Mann Whitney test. Percentage of lowering blood glucose levels of mature and immature aqueous corn silk extract was not significantly different. Immature aqueous corn silk extract was lowering blood glucose since 90 minute after the glucose load by 44,55 ± 4,92%. Whereas mature aqueous corn silk extract was lowering blood glucose since 120 minute after the glucose load by 30,15 ± 7,25%.
