Viabilitas Sel Hasil Preservasi Suspensi dan Pelet Staphylococcus aureus Menggunakan Medium Base Gliserol

Abstract


The aims of storage (preservation) as to culture collection and conserve stocks of microorganisms. The aim of this research was to determine the viability of bacterial cells on different cryprotectant agent. Three kind of cryprotectant agent glycerol base medium were applied to evaluatethe ability of bacteria growth in Agar medium. Isolat were used namely Staphylococcus aureus was preserved in 10%, 30% and 50% at 40 C for 180 days Immediately after preserved the bacteria was tested the viability at day 30 until 180 during storage. The enumeration of samples taken from cultures ini Mueller Hinton Agar. The results of the Kruskal Wallis and One Way Anova suspension preservation tests showed the effective time was day 30 at a glycerol concentration of 30%, the cell viability was 1.5 x 106 CFU/ml and a maximum of 120 days, the cell viability was 1.4 x 106 CFU/ml. The One Way Anova test for the best preservation time for bacterial pellets is 30 days with a 50% glycerol concentration which has a viability of 1.4 x 106 CFU/ml. The results of the Kruskal Wallis test show that there is no real difference between preservation using suspension and pellets, however, suspension preservation has a higher average cell viability.
